Description

Description: This assay employs a two-site sandwich ELISA to quantitate FGFRL1 in samples. An antibody specific for FGFRL1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any FGFRL1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FGFRL1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FGFRL1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: FGFRL1 is a member of the fibroblast growth factor receptor (FGFR) family, where amino acid sequence is highly conserved between members and throughout evolution. FGFR family members differ from one another in their ligand affinities and tissue distribution. A full-length representative protein would consist of an extracellular region, composed of three immunoglobulin-like domains, a single hydrophobic membrane-spanning segment and a cytoplasmic tyrosine kinase domain. The extracellular portion of the protein interacts with fibroblast growth factors, setting in motion a cascade of downstream signals, ultimately influencing mitogenesis and differentiation.
